Hepatocellular carcinoma (HCC) is a major cause of cancer-related mortality worldwide. Current immunotherapies provide benefits to only a subset of patients. Epigenetic therapies can modify tumour immunity and improve antitumour responses. However, resistance and an immunosuppressive tumour microenvironment remain major challenges. Pyroptosis is an inflammatory form of programmed cell death that can stimulate antitumour immunity. The STAT1–GSDME pathway may promote pyroptosis and enhance immune activation. This study investigates pharmacological activation of STAT1–GSDME pyroptosis to strengthen epigenetic immunotherapy for HCC.
